Histochemical Studies on the Staining Properties of the Mucosubstances in the Matrices of the Tracheal Cartilage

Abstract
Comparative histochemical studies were done on the mucosubstances in the¢¥ matrices of the tracheal cartilage of children, human fetuses, monkey, dogs, cats, rabbits, guinea pigs, rats, domestic fowls and doves. The mucosubstances were stained with alcian blue (pH1. 0^-2.5), PAS, aldehyde fuchsin (pH 1. 7), colloidal iron and alcian blue (pH2. 5)- PAS reaction, and those were also stained with the above methods following pretreatments of methylation, saponification, methylation-saponification and hyaluronidase.
The results obtained were as follows:
1. The mucosubstances in lacunar capsule and territorial matrix of tracheal cartilage of human and animal materials contained large amounts of sulfated mucin and nonsulfated mucin and the propor¡©tion of both mucins was different. The mucosubstances in the former were more sulfated, while those in the latter were more nonsulfated.
2. The mucosubstances in the granular interterritorial matrix of human and animal were composed of sulfated and nonsulfated acid mucosubstance and neutral, the proportion of both mucosubstances was different. The mucosubstance in the granular matrix of the human, monkey, dog and cat contained more sulfated and that of rabbit, guinea pig and rat contained nearly equal amount of both mucins. That of fowl and dove was more neutral.
3. The mucosubstances in the agranular and suhperichondrial matrix of human and animal were composed of sulfated and nonsulfated acid mucosubstance and neutral. The proportion of both mucosubstances was different, the mucosubstance in the former contained more acid mucosubsta¡©nee, while that in the latter contained more neutral.
4. Alcian blue (pH2.5) following pretreatment of methylation failed to stain the mucosubstances in the matrices of human and animal, while the pretreatments of saponification, methylation-saponi¡©fication and hyaluronidase digestion did not give any effect on their stainability with alcian blue and alcian blue-PAS reaction.
